Much like with Hatred, that doesn't work.

And to add flame to the fire, the game will sometimes crash when you quit. I think that may be the steam overlay though.
Those images are EXACTLY how it would look like if you have not done the steps in the guide D-Man11 provided (or did them incorrectly somehow).
If you've tried it for Hatred too (which we all know that is required for) and it hasn't worked for both games, then I hate to break it to you but you almost certainly are making an error somewhere in the steps and there's not much we can do to help with that. I encourage you to read over and follow the instructions repeatedly until something jumps out at you that you've been missing.

Or NVidia inspector isn't working on my system because Windows 10. Can YOU confirm on your Windows 10 annivesary updated system (assuming you have one) that it is working on IS defense?

I'm on Windows 10 anniversary, and have no problems with Nvidia Inspector on any single game. I don't own IS Defense so I can't confirm with that specific game, but I did not have an issue with Hatred which you are also having the same problem with.
I used to do IT support for a bank, and trust me, I would run into this exact scenario all the time, where one person would have an issue and no one else would, and 99 times out of 100 it would be an error on their part. Of course, the vast majority of people would come up with every reason/excuse that something is not working the way it should rather than accepting that they may have made an error somewhere, and in the end I would remote connect to their computer, find out what they did and show them.
Don't get me wrong here. I'm not trying to be an ass to you and just blame you for making a mistake, I'm actually trying to help you! I've been in the exact same situation myself on here once when I tried to follow Helifax's guide on here for setting up compatibility mode on a game. I swore up and down I did everything correctly, and he even asked me, "Are you sure you did ___?" and I was sure I did, but in the end he was right and I did not properly do the exact thing that he was saying, and I felt silly after that. Just goes to show that someone like myself (an IT support guy and one of the guys who makes fixes around here) can fall into the same trap. Unfortunately, there's not really anything anyone can do to help you in this case, because the correct instructions have already been provided, it's simply a matter of you figuring out where you are probably going wrong (my guess is that you do not have DarkStarSword's custom names XML properly installed, or you're not using Nvidia Inspector 1.9.7.3), but if there's any specific part you're not sure of, then ask and we'll try to answer.

No point, a new fix (that doesn't need Nvidia inspector) was uploaded on the Hatred thread works on both Hatred and IS Defense.
Discussion over.

Well, just so you know, that Hatred fix update is actually doing this step for you and automatically updating the Nvidia profile in the exact manner that if you followed the guide correctly you would have done yourself... which is convenient that it solves your problem, but only proves my theory that you were not doing it correctly.
So, essentially you've been given a fish rather than learning how to catch one on your own, but whatever works. Discussion over.

Time to be more civil to people who are trying to help you out.
How would any of us be able to prove NVidia Inspector is working on YOUR computer?
You'll still need to use NVidia Inspector for some games until they are updated. Lots of games that rarely get updated. You'll want to solve this problem.
You are the only person we presently know who has a problem with NVidia Inspector.

Actually, nvidia inspector crashed on my Win 10 box the other day (and the exact same version had worked previously), so there might be something to this, but I haven't investigated what caused it yet.
2x Geforce GTX 980 in SLI provided by NVIDIA, i7 6700K 4GHz CPU, Asus 27" VG278HE 144Hz 3D Monitor, BenQ W1070 3D Projector, 120" Elite Screens YardMaster 2, 32GB Corsair DDR4 3200MHz RAM, Samsung 850 EVO 500G SSD, 4x750GB HDD in RAID5, Gigabyte Z170X-Gaming 7 Motherboard, Corsair Obsidian 750D Airflow Edition Case, Corsair RM850i PSU, HTC Vive, Win 10 64bit
The crash in nvidia inspector is:
An unhandled exception of type 'System.IO.FileNotFoundException' occurred in mscorlib.dll
Additional information: Could not find file 'C:\WINDOWS\system32\nvd3dumx.dll'.
That is one of the files it checks to find some of the stereo settings names, and it appears that it is no longer present in recent drivers on Win 10 (my scripts pull that list of names from nvwgf2um.dll instead (Edit: which *also* seems to be missing in the latest Win 10 drivers)).

This issue has actually already been fixed in nvidia profile inspector 2.1.2.15. Please make sure you are always using the latest version.
The latest official release is 2.1.3.0 here:
https://ci.appveyor.com/project/Orbmu2k/nvidiaprofileinspector/build/2.1.3.0/artifacts
The latest continuous integration build (pre-release / beta) is always available here:
https://ci.appveyor.com/project/Orbmu2k/nvidiaprofileinspector/build/artifacts
Note that without that DLL, many of the stereo settings will not have names (except for some common ones I submitted to the project previously).

It's also possible to swap/import new profiles via GeForce 3D Profile Manager. OP can forum search it.
Perhaps OP could list the version of Inspector that he is using and compare version numbers to that of other W10 users that have it working.
Also, OP should know that not all profiles work with every game. If you are swapping complete profiles, in some games, the change "will not" take place until the game is restarted. Swapping a profile successfully will result in the overlay showing the name of the new profile. If the new overlay isn't seen, it's probably because that profile will not work with that game due to certain flags being present or missing.
