Have you tried doing a clean install of latest drivers? If so, how about some more details so maybe someone can help you better, like OS and GPU.

Try downloading them manually instead of updating, and do a clean install. http://www.nvidia.com/Download/index.aspx?lang=en-us

I don't know what to tell you as I'm not really sure what the problem is, if it was mine I'd start trouble shooting things ... it's odd that it was working fine and just stopped working, could be a software issue or a hardware one it's really hard to tell. You could try uninstalling all the NVIDIA drivers ... maybe even use something like DriverSweeper, you could try reseating everything, cables, GPU, monitor, etc ... personally I've had a windows update that botched everything on me, but I was able to just reinstall the drivers from NVIDIA.

Install them in safe mode after running the driver remover (also in safe mode, restart between the two)
