Having V-Sync on will slow down your FPS, but will help get rid of horizontal tearing which is common among 60 Hz refresh rate LCD monitors. With the new 120 Hz monitors, I am not seeing as much Horizontal tearing with V-sync off, so I have been turning it off to get more FPS. It is really up to you though, if you don't mind horizontal tearing and want more FPS, go with V-sync off. Otherwise, if horizontal tearing is making your game look bad, turn it on. Each game engine is different, so you will have to make adjustments on a game to game basis.

It might add some small delay, but you get rid of screen tearing.
I can only assume that tearing can even add some weird effects in S3D, as still don't own my S3D setup... yet :D

But, the point is - if screen can render for example 60hz, removing V-Sync and having '200 FPS' ingame will just mean within one frame that is drawn you'll have 2 'tearing' stripes -> as screen can not draw as fast as GFX card is drawing.

However, for super-human FPS players, V-Sync off might help a bit, because V-Sync (as I already said) does introduce some small delay.
But, for most people few ms (be it 5 or 30ms) is un-noticeable.
